abstract

results the pcr approved the bacillus genus of the isolates. the strain of bacillus subtilis was identified using biochemical tests. 40 ºc and ph 11 are the optimum condition for maximum keratinase enzyme activity. objectives the aim of this study was the isolation of feather degrading bacillus spp.from a poultry waste and the optimization of conditions for the highest enzyme activity and feather degradation. materials and methods the microorganisms were isolated from the waste of a poultry in miyaneh, iran, and the bacillus spp. were identified using morphological, physiological and biochemical tests. the bacillus spp. cultured in a medium consisted of feather at ph 7.4 and 27 ºc for seven days to identify the feather-degrading bacillus spp. the biochemical tests were performed to determine the strain of the bacterium. the study was repeated under different ph and temperatures to find the optimum conditions for best enzyme activity. background the feather is an environmental pollutant that can be degraded by bacterial and fungal microorganisms. the keratin sheets constitute 90% of the feather mass. due to the extremely rigid structure, keratin is insoluble and hard to degrade. some microorganisms such as bacillus spp. were reported to be able to degrade keratin by secretion of keratinase. conclusions b. subtilis was found to be able to degrade the feather.

Optimization of Culture Conditions for Keratinase Production in Streptomyces Sp. JRS19 for Chick Feather Wastes Degradation

The aim of this study was to find the optimal conditions of a newly isolated Streptomyces strain JRS 19. Screening for keratinase producing bacteria and their keratinase production were investigated. The keratinase produced by the isolate was purified by chromatographic techniques and the optimum pH maintained was 7.0 and maintained at different temperatures Viz., 20ºC, 25ºC, 30ºC, 35ºC and 40º...

Efficient Degradation of Feather by Keratinase Producing Bacillus sp.

Keratinase producing microorganisms are being increasingly utilized for degradation and recycling of poultry feather waste. Two native strains BF11 (Bacillus subtilis) and BF21 (Bacillus cereus) degrading keratin completely were characterized. The native strains produced more than 10 KU/mL of enzyme. Strain improvement resulted in isolation of MBF11 and MBF21 from BF11 and BF21 isolates, respec...

Screening of fungi efficient in feather degradation and keratinase production

Two hundred thirty four fungal strains were isolated by baiting method, for their feather degradation and keratinase producing ability. Fungi were tested on solid milk agar plates and in submerged culture. Maximum clearing zone was made by Chrysosporium indicum (7mm) on solid agar plates. The highest keratinase production was found in case of Acremonium strictum (74.40Unit/mL & 124.72 Unit/ml i...

Optimization for Keratinase Enzyme Production Using Bacillus thuringiensis TS2

Keratinase producing bacterial strains were isolated from the chicken feather dumping site. Keratinase producing bacterial strains were identified by physical, biochemical characteristics, fatty acid methyl ester (FAME) and 16S rDNA sequences. Medium optimization of the selected keratinase producing bacterial strain (Bacillus thuringiensis TS2) was checked with various parameters such as incuba...

Keratinase Production by Three Bacillus spp. Using Feather Meal and Whole Feather as Substrate in a Submerged Fermentation

Three Bacillus species (B. subtilis LFB-FIOCRUZ 1270, B. subtilis LFB-FIOCRUZ 1273, and B. licheniformis LFB-FIOCRUZ 1274), isolated from the poultry industry, were evaluated for keratinase production using feathers or feather meal as the sole carbon and nitrogen sources in a submerged fermentation. The three Bacillus spp. produced extracellular keratinases and peptidases after 7 days. Feather ...
